This insertion also results in expression of the thnL terminator in the host strain.

Using this system, we cloned the metagenomic DNA from an environmental sample collected from the shore of Punta San Garcia in Cadiz contaminated by crude oil from a tanker spill. The metagenomic DNA was transcribed by the T7 RNA polymerase from a vector containing the T7 gene 10 promoter, and its synthesis was terminated at the thnL terminator by the induction of nahR by salicylate in the host strain. Expression of the gfp gene was then monitored by fluorescent microscopy, revealing that transcription from the psal promoter is activated by NahR in a non-hosting strain and that synthesis of the N protein is able to impose processive anti-termination on nahR-induced transcripts.

We then tested the ability of this system to transfer metagenomic clones from the donor host to non-hosting strains by conjugation. To this end, triparental matings were performed with the rifampicin resistant and nalidixic acid resistant MPO553 and MPO555 carrying the bulk metagenomic library as donors, each of the above specialized host strains bearing pCC1FOS-CeuI and the metagenomic clones, and DH5a containing pRK2013 as the helper strain. We found that pMPO579 was transferred to all recipient strains with a very high conjugation frequency of over 10%, similar to that obtained with the mobilizable plasmid pBBR1MCS-3.

The ability to efficiently clone metagenomic DNA and to identify active clones is a critical step towards the effective utilization of the extraordinary biodiversity of uncultivated bacteria. However, a significant limitation of current functional metagenomic screening systems is the inability to detect complex regulatory systems by identifying their transcriptional activators or anti-termination proteins. The MPO555 system overcomes this limitation by integrating a promoterless GFP gene downstream of vector and metagenomic DNA cloning sites. This allows for exploitation of Substrate Induced Gene Expression (SIGEX) technology24 to identify regulatory systems by detection of GFP fluorescence, and by subsequent phage transcriptional anti-termination protein N-dependent processing of the cloned metagenomic DNA.

To develop the MPO555 system, a DNA fragment containing the lacUV5 promoter, the attenuator nasF and the thnL terminator of the lambda phage gene N was inserted into the E. coli EPI300-T1 strain at its trg locus, to yield pMPO571. This was then combined with the metagenomic DNA cloning site and the T7 gene 10 promoter to yield pMPO579. An additional modification of pMPO571 to yield pMPO580 involved cloning the thnL terminator upstream of a strong ribosome binding site from the T7 gene 10. The resulting system has the potential to identify regulatory systems by detection of T7 RNA polymerase-dependent gfp gene transcription in vivo.

Induction of a large number of transconjugants resistant to 100 mg L-1 carbenicillin (CbR) by the MPO555 system was demonstrated. The specialized MPO555 strains MPO554 and MPO555, which contain the metagenomic DNA cloning fragment and the activator NahR in their trg loci respectively, showed a 6-fold increase in CbR resistant transconjugants compared to the conventional EPI300-T1 NalR strain.

Detailed analysis of MPO555 strains showed that the psal promoter in pMPO580 is able to activate transcription from metagenomic DNA and that the thnL terminator can be evaded by NahR-dependent processive anti-termination mediated by a truncated lambda phage N gene. However, induction of gfp expression from psal in the presence of arabinose and salicylate was very low. Increasing the plasmid copy number resulted in a modest increase in gfp expression, but high levels of gfp expression were obtained when arabinose and salicylate were added together. This demonstrated that the thnL termination signal is not essential for activation of gfp transcription in this system and that production of NahR and N can significantly improve the performance of the MPO555 system.

Batman 138 Review

 

The Gotham War continues, and this time it’s a war within the Bat Family. Father versus son, teacher versus student, this internal conflict is sparked by a shocking betrayal. It may not be as big as a clash between Catwoman and Batman, but this brawl within the Bat-family does get a lot of attention, and for good reason.

This issue sees the devolution of the entire Bat Family, with some of the most brutal consequences we’ve seen yet in this event. It’s not easy to write about this kind of story, but Chip Zdarsky and Jorge Jimeneze do an exceptional job of it here. They capture the characters in a very raw state, with scars and bruises and burnt skin marking their weariness. The art also has a very rough and gritty look to it, with each action sequence extending over large areas of the page.

The story begins with Batman and the other Robins hunting Simple Simon, who attempts a third heist at a winter resort, smashing a hotel into the ice and stealing an ice boat. The other Robins catch him, but the action gets stale when the entire scene ends with a fight between Bruce and Jason that isn’t very interesting at all. The rest of the issue sees the other Robins fighting each other, with the final scene involving Damian being towed away by Tim Drake, presumably to stop him from killing his dad.

One of the most interesting aspects of this arc has been watching how each character has become more unhinged. This is especially true of Batman138, who seems to have lost all sense of right and wrong as the saga has progressed. He’s starting to do things that would be completely out of character for any normal person, and it’s pretty scary to watch.

In the end, the Gotham War reaches its climax, with Vandal Savage appearing and taking over the world. It’s not the most original premise in the history of comics, but it’s still an entertaining and exciting story that has some very cool moments. This is one of the best events that DC has put out this year, and it’s definitely worth checking out!
